Das Gebiet von Montefalco war bereits in der Römerzeit ein beliebter Ort für die Adligen der Gegend, die dort ihre Villen bauten. Im frühen Mittelalter taucht die Stadt in den Texten mit dem Namen Coccorone (noch nicht eindeutiger Ortsname) auf und wurde erst im 13. Jahrhundert von Friedrich II., der von der großen Zahl der Falken in der Region betroffen war, in Montefalco umbenannt. Die Stadt (seit 1280 Todi unterworfen) wurde zur Gemeinde mit dem Statut von 1282 erklärt. Unter der Besetzung von Todi begann der Anbau der Rebsorte Grechetto in der Gegend, die die bereits rentable Rotweinproduktion ergänzte. Die Stadt trat in den Bereich von Spoleto und wurde als Sitz der Herzogtumsrektoren (1320 – 1355) ausgewählt und ging dann in den Besitz der Famile Trinci von Foligno über. Die von Giovanni Vitelleschi im Namen des Papsttums wiedergewonnene Stadt blieb bis zur Einheit Italiens Teil des Kirchenstaates.

Derzeit ist das Städtchen hauptsächlich für die Herstellung des hervorragenden Sagrantino di Montefalco DOCG (monovarietaler Wein, dessen erstes schriftliches Zeugnis im 16. Jahrhundert erschien) und des ähnlichen Rosso di Montefalco DOC bekannt.
